Cleavage Activation of the Human - Adapted Influenza Virus Subtypes by Kallikrein - Related Peptidases
نویسندگان
چکیده
Background: Cleavage of the influenza hemagglutinin (HA) precursor by host proteases is a critical step in the virus life cycle. Results: The airway, secreted proteases kallikrein 5 and 12 were found to cleave and activate influenza HA. Conclusion: Each peptidase had distinct preferences for particular human-adapted, influenza viruses. Significance: Identification of these HAcleaving peptidases aids in our understanding of host proteases involved in influenza infection.

A peptide-based approach to evaluate the adaptability of influenza A virus to humans based on its hemagglutinin proteolytic cleavage site
Cleavage activation of the hemagglutinin (HA) protein by host proteases is a crucial step in the infection process of influenza A viruses (IAV). However, IAV exists in eighteen different HA subtypes in nature and their cleavage sites vary considerably. There is uncertainty regarding which specific proteases activate a given HA in the human respiratory tract. Understanding the relationship betwe...

Abstract : Avian influenza virus (AIV) infection is a major cause of influenza mortality in birds and can cause human mortality and morbidity. Although the risk of infection with avian influenza virus (AIV) is generally low for most people, the pathogenic virus can cross the species barrier and acquires the ability to infect and be transmitted among the human population; therefore the ra...

Background and Aims: The caspases are unique proteases that mediate the host cell apoptosis during viral infection. In this study, we identified the caspase cleavage motifs of H5N1 and H9N2 influenza viruses isolated during 1998-2012. Materials and Methods: Amino acid sequences of the eleven proteins encoded by the viruses as the caspase substrates downloaded from NCBI. The caspase cleavage mot...
متن کاملKallistatin ameliorates influenza virus pathogenesis by inhibition of kallikrein-related peptidase 1-mediated cleavage of viral hemagglutinin.
Proteolytic cleavage of the hemagglutinin (HA) of influenza virus by host trypsin-like proteases is required for viral infectivity. Some serine proteases are capable of cleaving influenza virus HA, whereas some serine protease inhibitors (serpins) inhibit the HA cleavage in various cell types. Kallikrein-related peptidase 1 (KLK1, also known as tissue kallikrein) is a widely distributed serine ...
متن کاملHeamagglutinin Conserved Domain (HA2) Prepared in Prokaryotic System is Immunogenic in Mice but not Protective against Lethal Influenza Challenge
Background and Aims: Influenza vaccine production process is time-consuming with little-to-no cross-protection which requires annual adjustment. The construction of a universal vaccine to deal with the pandemics and epidemics which occasionally threat human population is the aim of many researches worldwide. Today, influenza vaccines are mostly against two major antigenic proteins, hemagglutini...
